The American Information Bureau has created a team to hunt down and destroy Nightwalkers - modern day vampires. The player controls John Lloyd, an agent on such a team, sent in to discover the whereabouts of an advance recon team that has gone missing in a city swarming with Nightwalkers. It's up to John and his team to hunt down the Prime Walker and clear the city of the vampire infestation.

Vampire Rain is a third-person shooter game where stealth play is key, similar to games like Splinter Cell and Metal Gear Solid. The gameplay involves a lot of platforming, exploration, and combat-style elements. Players often need to use the darkness to stay undetected during missions. The arsenal includes various firearms such as a handgun, sub-machine gun, assault rifle, shotgun, sniper rifle, anti-armor rifle, and a UV Knife. Traps are more effective against exceptionally strong enemies, and killing the Prime Walker allows players to take out large groups of Nightwalkers at once.
