The game PlayStation Move Heroes features both single-player and two-player co-operative multiplayer modes, where players explore levels in a style reminiscent of Super Mario 64. The game boasts six playable characters: Ratchet, Clank, Jak, Daxter, Sly Cooper, and Bentley. A total of 50 levels are available, categorized into five "archetypes", five weapon classes, and four environments. Players can choose which character to play as on each level, with each character having a unique gameplay style and advantages on certain levels. Co-operative play allows the second player to automatically select the sidekick character based on the first player's choice.

The game's five weapon classes are: Disc, Whip, Bowling, Projectile, and Melee. The disc is thrown like a frisbee and navigated using the Move controller to achieve objectives, such as hitting moving targets. The whip is controlled via 1-to-1 movement with the Move controller. Bowling involves throwing the bowling ball and guiding it with the Move controller. Projectiles involve using guns and other weapons, such as grenade launchers. Melee utilizes each character's iconic weapons to hit enemies in a similar manner to their original games.
