Earth Defense is set in a world where aliens have invaded Earth, prompting the United Nations to create a flying craft called the "Phoenix Spitfire" to fight back. The player takes on the role of the pilot, tasked with defending the planet from enemy attacks.
The gameplay involves navigating through five vertically-scrolling levels, targeting enemy aircraft and facing off against bosses at the end of each level. Along the way, players can collect power-ups dropped by defeated enemies to upgrade their cannon, adding new abilities such as the Tri-Shot, Napalm Bomb, Rear Shots, and support drones.
