de Blob 2's story begins with Prisma City , which is facing an election that could change its fate. A dark figure named Papa Blanc devious plans to rig the elections and take power, but it turns out he's working for a sinister corporation called The INKT Corporation. As he succeeds in draining colour from the city, Blob must journey through Prisma City to restore its vibrant colours and save its inhabitants. His mission is to stop Papa Blanc, free the population, and defeat The INKT Corporation once again.

The gameplay of de Blob 2 has been updated for a more standardised console control scheme on Nintendo Switch, unlike the original. Players still change colours by absorbing paint, but now they also enjoy platforming and sliding gameplay as they paint the world with various hues. The game features a competitive multiplayer mode where two Blobs compete to see who can paint the most and collect points first. The character Pinky provides assistance in cooperative play.

Compared to the previous title, de Blob 2 has more types of enemies, power-ups, and higher resolution graphics. Blob has a new dash attack that costs points, as well as upgrades like better defence and increased paint carrying capacity. Power-ups also provide various benefits, such as becoming a wrecking ball or turning into a black hole. The game places greater emphasis on platforming challenges, where Blob must complete tasks to unlock buildings and progress through the story.
