The gameplay of NSMBU features tight platforming and power-ups such as mushrooms, fire flowers and stars. Players control Mario or Luigi through various levels, each with its own unique theme and challenges.

Additionally to the core campaign, there are also challenge modes that ask players to complete certain tasks and boost rush and coin battle modes that add variety to the gameplay.
