Lego Marvel Super Heroes is an action game that brings together over 100 Marvel heroes and villains from various franchises, with the player controlling the good ones to save Earth from a team of villains led by Galactus. The game features a blocky cartoonish visual style and gameplay reminiscent of LEGO adventures.

The game world consists of Manhattan as an explorable open world, including known Marvel locales such as the Marvel HQ, and a series of self-standing story mode levels located in different worlds, including Asgard, Bifrost, and Asteroid M. Players can explore Marvel-related buildings in New York and hear narrated storylines by Deadpool.

At the start of the game, players control Iron Man and Hulk, soon joined by Spider-Man. As they progress, more characters are unlocked by advancing through the game or collecting Gold Bricks found while exploring Manhattan, completing side quests, and completing story mode levels. Extra outfits and vehicles can be obtained by retrieving Character tokens and Vehicle tokens in Manhattan. Other collectibles include Minikits, Deadpool bricks, which provide power-ups and extra features.

The game has a large cast of 27 voice-acting actors for the in-game dialogues. On the Nintendo Switch, players can enjoy the game without a TV using its GamePad's off-tv play feature. In two-player mode, one player uses the display on the GamePad, while the other avoids split-screen gameplay.
